Elamipretide (SS-31) is a synthetic tetrapeptide that is mitochondrial-targeted. The peptide is a four-amino-acid and it can reach the inner membrane of mitochondria easily. SS -31 attaches itself to cardiolipin in laboratory studies – a major phospholipid that assists in maintaining the shape and proper functioning of mitochondria. This binding stabilizes the membrane, lowers reactive oxygen species (ROS), and shields integrated mitochondria, amplifying the energy production by the cell. In cell and preclinical research, SS -31 is ligand-bound to investigate mitochondrial dysfunction, activate protective cellular responses, and understand the pathways of oxidative stress. Molecular Structure and Composition

SS31 (Elamipretide) is a synthetic tetrapeptide that is made up of D-arginine, 2,6-dimethyltyrosine (Dmt), Lysine, and Phenylalanine, with its C-terminus aminated. It has the order D-Arg-Dmt-Lys-Phe-NH 2. Methylated dimethylated tyrosine and additions of a D-amino acid are found to improve enzyme resistance to eventual degradation, and also improve its uptake in mitochondria during laboratory experiments. The molecular formula is C₃₂H₄₉N₉O₅, and it has a molecular weight of about 639.8 daltons. To study the structure and functioning of mitochondria, it can be easily accumulated in the inner membrane of the mitochondrion, which is why it is also an excellent instrument because of its small size and positive charge.

Mechanism of Action

Since it is positively charged and small in size, SS-31 is rapidly absorbed in cells and accumulates in the inner mitochondrial membrane. It targets phospholipid cardiolipin, which holds the cristae of the mitochondria and helps the electron transport chain. This binding accelerates the speed of electron flow, reduces the generation of ROS, and increases ATP-generation.
